Dishes and glassware
THICK PLASTIC AND GLASS ONLY IN THE AUTOCLAVE. Thin plastic such as yellow graduated cylinders bumper and Tupperware will melt.
- Load and run dishes in dishwasher
- Rinse dishes if necessary/make sure all solids are disposed of properly (agar gel, etc.)
- Follow instructions by the dishwasher
- Cycle time: 199 minutes
- Anything too large for the dishwasher should be hand-washed for the autoclave
- Wrap dishes with foil and attach a small piece of striped autoclave tape
- Cover open mouth containers
- Completely wrap utensils and flat objects
- Cap screw on bottles (should not be tight, just enough not to pull off)
- Load autoclave
- Place dishes in bin
- Slide bin into the middle of autoclave chamber
- Close door all the way and turn the wheel until the screen says “Closed”
- Run autoclave
- See instructions taped at the top of the autoclave
- Use Gravity 15-10 cycle (GR15-10 on the screen)
- Cycle time: 30 minutes
- Set a timer, as the autoclave must be emptied and closed immediately after each cycle so that it can reheat for any subsequent cycle
- Observe the autoclave after start in case any water is dripping from door, the door is not fully closed, or any other issue
- Unload autoclave
- Make sure screen says cycle is finished
- Open door, beware of steam
- The bin is hot; use orange heat gloves in the top drawer to the left of autoclave
- Close door all the way
- Click “No” when it asks to print a cycle report unless there is a malfunction
- Allow dishes to cool before putting them away
- Lab-owned items go into the cabinet with the other pink glassware or on the counter under that cabinet in the Molecular Suite
- Items not belonging to the lab go on the shelves in P376
Solid waste in biohazard bags
Biohazardous solid lab waste (such as paper towels, gloves, tubes, etc.) goes into orange biohazard bags
- Once an orange biohazard bag has been filled, hold it closed at the top (but do not tie it off), and take it to the autoclave room
- Place the bag into a bin and tape the top shut
- Use Gravity 15-1 cycle (GR15-1 on the screen)
- Cycle time: 25 minutes
- Follow the procedure above for the loading and unloading of the autoclave
- Once the bag has been removed from the autoclave, it can be thrown away in the regular trash after cooling
Liquid Waste
Any waste liquids that contain organic waste should be dumped and rinsed into large waste beakers or kept in their original containers if applicable.
- After the conclusion of experiments, place vessels with liquid/organic waste in a sink
- Pour a generous amount of germicidal bleach into the vessel (can stop pouring once the liquid in the vessel changes color)
- Leave the bleached waste overnight
- The next day, pour and rinse out the vessel down the sink
